The insert contains internal HindIII and AccI sites. Restriction digests of the clone give the following sizes (kb): BamHI--2.9, 0.90; EcoRI--3.8; PstI--3.8; HindIII--3.0, 0.87; XbaI--3.8. Linearize the plasmid with AvaI for in vitro transcription. This is a full-length clone of virus segment S12. The cDNA ends were tailored to terminal BamHI sites before cloning into the BamHI site of pGEM-1. |
